If you are looking for best Rune DPS for PVE content, you should use Rage+Blade Runes.

1. First. this Rune you should find substat that have atk%, crit dmg%, crit rate%. And for the last substat I prefer get hp% or hp or acc%.
2. 2nd. this Rune find atk% main stat and with substat that have crit dmg%, crit rate%. And for the rest 2 options is hp% or hp or acc% or def% or def.
3. 3rd. this Rune you should find substat that have crit dmg%, crit rate%. And for the rest 2 options is hp% or hp or acc% or def% or def.
4. 4th. this Rune find crit dmg% main stat and with substat that have atk% and crit rate%. And for the rest 2 options is hp% or hp or acc% or def% or def.
5. 5th. this Rune you should find substat that have atk%, crit dmg%, crit rate%. And for the last substat I prefer get hp% or hp or acc%.
6. 6th. this Rune find atk% main stat and with substat that have crit dmg%, crit rate%. And for the rest 2 options is hp% or hp or acc% or def% or def.

And if you want to use for pvp content : 

2nd and 6th can replace with hp% main stat and with substat that have atk%, crit dmg%, crit rate%. And for the last substat I prefer get def% or def or acc%.

Note: 4th Rune can switch to crit rate% main stat if you need more crit rate%. 

For enhancement, each Runes should have min. S3 substat for one stat. Example : First Rune with substat that have atk%, crit dmg%, crit rate%. Find min. S3 for atk%.

After your Runes have a good stats then you can apply effect and enchant using atk% / crit rate% / crit dmg% depends on what stat that you are lacking. If your stat lack of crit rate% and atk% then apply effect and enchant to your Runes crit rate% and atk% stat.

Get Your crit rate like 90% minimum, Crit dmg 270% minimum and 8500 Attack for your Perna. That is just one of example Rune stat.
